Okay, I am 16 I have a dilemma, I like this girl alot, but I don't really know if she is interested. Well I have been single for around a year now, and I really want to get back into a relationship, I want to know what I should do in order to get her to like me back. I haven't done this in a while so I am rusty, if anyone can help that would be great, thanks!!

Are you friends with her now? Do you two talk now? I would advise you to start talking to her, get to know her. Don't rush into a relationship, you two need to know one another before that.
You shouldn't "try" and make a girl like you, if they feel a connection, then they will like you. If they think that you two will make better friends, then they won't like you.
Hmm, you want to get back into a relationship? Are you sure you want to be in a relationship with this girl? Or just any girl? Maybe you should think about that.
If you want to be in a relationship with her, and not just in a relationship in general, then talk to her. Call her, text her, IM her. Sit with her at lunch, anything. Be nice to her, listen to what she has to say. Don't try to act all cool and show off.

I agree with Autumn here you really should think about if you want to be with this girl because you have strong feelings for here, or you just want a girlfriend. It's pretty common for guys to think they like a girl and say they like them and try to get with them just so they can have a relationship and do stuff. The only girls that you should go out with/want to go out with are the ones you really like because of there personalities or other things besides appearance that you are attracted to. You will know when its right, trust me, don't force anything.

We have been friends for a while now, I really like her, I don't just want a girlfriend for a relationship.
