In theory, yes- although it won't necessarily work that way. When you reproduce, or trick your body into thinking you are, it releases a heap load of various chemicals and hormones. One of these is called serotonin. This is currently being investigated although scientists believe the quantity of this is linked to headaches and migraines. They believe that having higher levels of serotonin decreases the pain of the headache. As sex or masturbation releases serotonin, it would theoretically help with headaches.

Masturbating is a temporary solution to curing a headache. While you're masturbating, yeah it feels super good and so for the time being, the pleasure overcomes the headache.

But once you're done, there's no more pleasure, so the headache can be felt again, but it'll feel worse.

You see, the reason for headaches is constricted blood vessels in your head. Masturbating or having sex opens up those vessels and so the headache is gone. But once you stop, the vessels constrict again causing it to feel worse for you.

Depending on the cause of the headache the increase in tension prior to orgasm can temporarily make it worse. At orgasm the body release endorphins which act as naturally pains killers so this may well make the headache disappear. This effect is also temporary, though probably lasts a little long than the pre-orgasm tension.

Given the temporary nature of the relief and the soporific effect of orgasm this would be an effective solution if the headache is preventing you from going to sleep, i.e. masturbate and then drop off to sleep in the afterglow. If the problem is at another time of the day you'd be better drinking some water, taking an over the counter painkiller and trying to concentrate on something else.

Scientifically, yes. Orgasm releases endorphins such as dopamine into the body (dopamine is sort of like the body's morphine) and will help lighten your headache. Now whenever your girlfriend uses a headache as an excuse not to have sex, you can tell her about dopamine.
